应用ELISA试剂盒检测鱼肉中孔雀石绿和结晶紫

    Development of enzyme-linked immune-sorbent assay for detection of malachite green and crystal violet residue in fish

    • 摘要: 开发了一种用于检测鱼肉中孔雀石绿(MG)和结晶紫的间接竞争酶联免疫(ELISA)检测试剂盒,并对试剂盒性能进行了验证。结果表明,MG浓度在0.2~5.4ng/mL范围内有较好线性关系,50%竞争抑制率为0.50ng/mL,试剂盒检测鱼肉中MG的最低检测限为0.17ng/g。鱼肉中分别添加0.2、0.4、0.8 ng/g的MG标准品,回收率均在72.4%以上,批内变异系数小于12.4%,批间变异系数小于12.9%。本方法灵敏度高,重复性好,检测时间短,可同时检测鱼肉中孔雀石绿、结晶紫、隐性孔雀石绿、隐性结晶紫等四种药物的残留总量。

       

      Abstract: An enzyme-linked immune-sorbent assay (ELISA) method was developed to determine the malachite green (MG) and crystal violet residues in fish and the performance of the ELISA method was evaluated. The results showed that the ELISA kit exhibited a good linearity in the range of 0.2 ng/mL to 5.4 ng/mL, and the 50% competitive inhibition rate was 0.50 ng/mL. The low limit of detection for MG in fish muscle was 0.17 ng/g. For the spiked experiment, the average recovery rate was higher than 72.4%, the coefficient of variations (CV) for intra-assay and inter-assay were lower than 12.4% and 12.9%, respectively. The developed method was sensitive, accurate, rapid and suitable for the determination of malachite green, crystal violet, leucomalachite green and leucocrystal violet total residues in fish.
